CHRNA7 - nicotinic acetylcholine receptors

GENE DESCRIPTION:

  • Function: CHRNA7 is a nicotinic acetylcholine receptor acting as a ligand-gated ion channel for mediating fast signal transmission at synapses.
  • Disease: Genetic defects in CHRNA7 have been found to be highly associated with Autistic Disorder.

rediMODEL-KO

CHRNA7 RediMODEL KO: Generated in the C. elegans homolog eat-2 with an early stop in coding sequence (ad465) which create a knock-out (KO) strain. Loss of function from eat-2 deletion leads to sluggish uncoordinated activity. Major phenotype readout is the observation of 75% reduction in pumping rate. An outcrossed version of the early stop mutant (ad465) is currently available for preorder as gene knock-out.

 

CHRNA7 RediMODEL KI: Generated in the C. elegans homolog eat-2 with human gene insertion to create a knock-in (KI) strain. Entire coding of eat-2 is replaced with wild-type CHRNA7 coding sequence. A CHRNA7 gene replacement is available for preorder as a humanized wt line.

 

CHRNA7 RediMODEL CV: Generated in the C. elegans homolog eat-2 with human gene insertion containing a clinical variant allele to create a clinical-variant (CV) strain. Entire coding of eat-2 is replaced with a CHRNA7 coding sequence containing an amino-acid change polymorphism that has been observed in patient populations. Upon request, a CHRNA7 line with client-specific polymorphism is made as a clinical variant allele.

KNOCK-OUT of C. elegans gene:

eat-2 is a homolog gene to CHRNA7. The eat-2 gene functions to control neurotransmission across the neuromuscular junction. When eat-2 is expressed and active, it gates electrical signals that control neurological activation of the pharynx. Deletion mutation in eat-2 causes a loss-of-function phenotype as a reduction in pumping rate. The canonical allele in eat-2 is (ad465), which contains an early stop in the gene coding region and is thought to render a knock-out (KO) of gene function. The most easily measured phenotype for the KO is a pharynx pumping loss-of-function phenotype. To decrease the risk of background mutations, the KO strain is 4x outcrossed. The resulting outcrossed strain containing (ad465) allele is offered as the CHRNA7 RediMODEL-KO strain. The KO strain is confirmed by PCR and shipped in the RediMODEL kit format.  Included in the kit is N2 wild-type strain as a reference control.

Each kit arrives with 3 frozen tubes of CHRNA7 RediMODEL-KO and 3 tubes of N2 for control and all materials needed for 2+ days of growth.

CLINICAL VARIANT C. elegans:

eat-2 is a homolog gene to CHRNA7. The eat-2 gene functions to control neurotransmission across the neuromuscular junction. When eat-2 is expressed and active, it gates electrical signals that control neurological activation of the pharynx. Deletion mutation in eat-2 causes a loss-of-function phenotype as a reduction in pumping rate.  Installation of a CHRNA7 in replacement of eat-2  results in the CHRNA7 RediMODEL-KI control strain and cause restoration of normal pharynx pumping phenotype.  A client-specified clinical variant is installed into the CHRNA7 sequence of the wt-control strain.  The result is a CHRNA7 RediMODEL-CV strain whose relative phenotypic consequence can be rapidly measured. The CV strain is confirmed by PCR and shipped in the RediMODEL kit format. Included in the kit are CHRNA7 RediMODEL-KO and CHRNA7 RediMODEL-KI as reference controls.
