RecombiREAD Reagent. Experiments in C elegans demonstrate recombiREAD reagent can act as co-CRISPR tool that acts within 24 hrs of injection.
RecombiREAD-RED Reagent designed to turn red in C. elegans embryo after injection. The recombiREAD reagent has significant utility act as co-CRISPR tool that acts within 24 hrs of injection.
Using a RecombiREAD reporter: "reporter RED", examination of embryos at 24 hrs for high levels of red embryos provides indication the remaining cohort has high likelihood of containing a desired edit. For example, using the RecombiREAD reporter reporter RED as a co-CRISPR reagent to detect dpy-10 edits as roll-positive animals, 76% of plates marked by presence of red embryos had desired CRISPR edit. Contrast to a plasmid contain a RFP driven by constitutive promoter, only 21% of of marked plates containing desired edit. Without selection marker, less than 10% of all plates had desired edit.
Currently RecombiREAD reporter reporter RED is available for evaluation. Kits of RecombiREAD are planned to be available in additional fluorophore markers.
